From 2016 under supervision of Dr. Tatsuo Fukagawa in Osaka University, Japan, I started to work on the regulation of the Kinetochore-centromere interaction in Chicken DT40 cells and Human RPE1 cells and in vitro binding assay. Recently I revealed that one of the kinetochore component CENP-C changes the interaction with the CENP-A nucleosome during cell cycle progression. This change is regulated by CDK1 phosphorylation in cells and in vitro. I reported its regulation and biological significance in DT40 cells and RPE1 cells (Watanabe et al., J. Cell Biol. 2019, Watanabe et al., Bio-protocol 2021).

I’m working on molecular dynamics of the kinetochore proteins during the cell cycle progression with FRAP assay in multicolor live-cell imaging.

In parallel, I’m analyzing M18BP1/KNL2 that is an essential protein for maintaining CENP-A to the centromere region. KNL2 directly binds to CENP-A nucleosome through CENP-C like motif as same as CENP-C.

In Dr. Tatsuo Fukagawa Laboratory, I acquired the techniques of cell biology and biochemistry. For example, CRISPR/Cas9 genome editing, Immunostaining, Immunoblots, live-cell imaging for FRAP, in vitro protein purification, nucleosome reconstruction, and in vitro binding assay.
